Need plumbing help ASAP!!!!


If you have a hole/leak, and the pipe is not copper- it is likely because the pipe has rusted through, either from the outside in or inside out. In any case, if it has one hole it is likely the pipe wall is thin in other areas and another hole will appear sooner or later. Be careful if you use a clamp for a temporary fix you could collapse the pipe!

Black iron or galvanized steel pipe is not generally used any more for water pipe. The best recommendation is replace the entire length of pipe with copper. If you are not comfortable doing it yourself, call a plumber. He will shut the water off at the curb and should be able to do the necessary work in about an hour or less, especially if you have already exposed the pipe at the leak.

If the line from the street is iron or steel too, then you might consider replacing that as well. It is not a question if it will fail, but when it will fail.

Investing vs Gambling Meta-Post | Kid Dynamite's World

Please allow me to babble out loud (in pixels?) here for a minute? This won?t be the first nor the last time I make the comparison between the financial markets and casino gambling, but there are important distinctions that need to be elucidated.

One of my first exposures to financial markets was the interview process coming out of MIT.? I related some stories from interviews with Susquehanna in an earlier post.? ? In that same post, I made some analogies between sports betting and investing in CDOs.? Anyway, Susquehanna was by far the most ?gambling? oriented firm I ever spoke with.?? That doesn?t mean that they want their traders to be free wheeling, loose swinging, risk taking cocaine cowboys ? quite the opposite, in fact.? The crux of Susquehanna?s training program was to teach their new-hires that like poker, craps, blackjack and sports betting, trading and investing have a large ?luck? component too, and that you need to be aware of all of the factors that can alter the balance of luck and skill in the equation.

When you sit down at the poker table, you need to try to put yourself inside your opponents? heads.?? When you?re trading stocks, bonds, options, seashells, or anything else, you need to do the same (I?ve written about that before too ? in the form of avoiding confirmation bias).? When you walk into a casino and sit down at a Spanish 21 table, you need to understand the rules of the game and know what you don?t know, instead of saying ?holy cow ? no bust! this is awesome! free money!??? When you?re trading stocks, (achem, perhaps, TVIX?) you need to do the same.

That doesn?t mean, however, that you should treat your investing portfolio like your gambling portfolio.??? I mention this because I?ve seen a bunch of people write things lately like ?only invest what you can afford to lose.?? It?s not that this is necessarily terrible advice ? it?s more that this is a terrible ?investing? strategy:? that?s not how you invest, that?s how you gamble.?? When I plunk down $500 at the blackjack table at the Palazzo to play some green chip blackjack with The Big Show, I know that I could lose the whole buy-in.?? When I buy a stock position, however, I?m almost never expecting to be anywhere near the position of being able to lose my entire investment.?? That?s because I treat my ?investing? portfolio different from my trading portfolio.

There are certainly times where I am in a trading position that could lose 100% ? I wrote about one such time with Sino Forest last year.? I think what I?m trying to say here ? and yes, I realize I?m babbling (disclosure: I warned you in the first sentence!) ? is that when you?re in a ?only invest what you can afford to lose? situation, it?s obviously essential that you 1) realize that a total loss is a possibility (you probably already realized that with the mantra about ?only what you can afford to lose?), but more importantly, perhaps: 2) ask yourself why you?re putting yourself in such a ?gambling? position, what you think your edge is and why you?ll be able to come out on top.

I actually tried to explain my reasons for ?gambling? on Sino Forest in the comments of that prior post ? the gist of it was that even though I wasn?t an expert in Chinese Forestry, I didn?t think that most of the people trading the stock were experts either ? I thought that they were selling first and asking questions later,? and I thought it was an opportunity to buy panic for a trade.?? It turns out that I managed to escape this trade with a nice gain, even though I was wrong about Sino Forest in the end ? the stock was halted and hasn?t yet re-opened for trading.? The important thing is that I had my reasons for the trade, and they were never ?everyone is wrong about Sino Forest and I?m right even though I haven?t been to see all of their forestry operations for myself and I have no basis for belief that they are a long term value play.?? It was a trade (not a long term value investment), it worked (although it?s important for me to understand that I can?t be sure it worked for the reasons I thought it was going to work!), and it may have been very lucky that I escaped before the hot potato exploded.

Anyway, I don?t know if this post made any sense ? my point was to try to give some thoughts on the Venn-Diagram inside my head that represents investing, trading, gambling, and how they all interact.

Tea party leaves imprint on GOP convention

(AP) ? There's a marriage of convenience at the Republican National Convention this week.

Mitt Romney needs the tea party's vigor to ensure the GOP's conservative base is fired up and works on his behalf this fall. The three-year-old tea party movement needs the Republican Party and its new standard-bearer to champion its causes and get its agenda enacted. And the two sides have never had much affinity for the other.

Yet, the tea party has left a solid imprint on its first GOP presidential convention, from the speeches to the platform to the displays of deep opposition to President Barack Obama. With this backdrop, Romney ? hardly the first choice of this group of insurgents ? will accept the nomination Thursday.

"They've been very successful in leaving their mark at this convention," said Republican National Committee Vice Chairman Jim Bopp of Indiana. "They are a vital part of our coalition."

But Romney and the tea party have a difficult past.

The former Massachusetts governor, whose signature accomplishment was passage of a state health care overhaul that Obama's was modeled after and that the tea party despises, never warmed to the insurgent activists and resisted a full embrace of them.

As the GOP looked for a presidential candidate, the tea party rallied behind just about everyone for at least a time, from Minnesota Rep. Michele Bachmann to Georgia businessman Herman Cain to former House Speaker Newt Gingrich to former Pennsylvania Sen. Rick Santorum.

It's safe to say the tea party was, at best, resigned to Romney as its nominee when it got a welcome boost as the candidate chose Wisconsin Rep. Paul Ryan ? a favorite among this group ? as his running mate.

The tea party is more influential within the Republican Party than within the overall voter population.

According to a recent Associated Press-GfK poll, 48 percent of Republicans considered themselves supporters of the tea party movement. However, only 29 percent of all registered voters considered themselves tea party supporters, and less than 10 percent said they strongly support the movement.

If they turn out in force, these voters could be influential in competitive states such as Iowa, Michigan, Ohio and Wisconsin, where some conservatives have had misgivings about whether Romney is conservative enough.

With Ryan on the ticket, there was something for them ? and their preferred politicians ? to celebrate this week in Tampa.

"This national movement is fueled by what unites us: a love of liberty, a belief in the unlimited potential of free men and women," Ted Cruz, a tea party-backed U.S. Senate candidate from Texas, declared from the podium this week, igniting cheers from inside the Tampa Bay Times Forum.

While not everyone made such direct overtures to the tea party, its rhetoric of uncompromising spending cuts, strict constitutional adherence and limited government dotted speeches from the GOP's rising stars to the graybeards.

In attendance are some of the movement's heroes, including New Hampshire Sen. Kelly Ayotte and Florida Sen. Marco Rubio. Another favorite, Kentucky Sen. Rand Paul, was greeted with cheers of "Liberty!" when he stepped to the podium Wednesday.

"We have nothing to fear except our own unwillingness to defend what is naturally ours, our God-given rights," Paul said during his speech to the convention, prompting a renewed roar.

The insurgents' imprint went beyond the rhetoric.

Tea party activists also won changes to the Republican Party's platform, including nearly a dozen fiscal positions, such as studying returning the United States to the gold standard and requiring a two-thirds majority in Congress to pass a tax increase.

But there was tension behind the scenes when party officials sought to weaken tea party influence.

First, the RNC's rules committee voted that it could change the party rules in the middle of the four-year cycle ? instead of every four years, as has been the practice. This is a particularly sensitive point with the tea party's rule sticklers.

Another new rule was aimed at limiting the ability of insurgent presidential candidates to amass delegates to future Republican conventions. They require states to award delegates to candidates based on the outcome of primaries and caucuses.

The new rules originally allowed the likely presidential nominee to choose which delegates would represent them at the convention ? taking that power from state parties, which prompted sharp opposition from tea party activists. In a concession, party leaders agreed to remove the language, instead saying delegates who support candidates other than the one they are obligated to support shall have their votes nullified.

The change appeased some state party activists who didn't like the idea of candidates mandating which delegates could attend the national convention. But it didn't satisfy supporters of Texas Rep. Ron Paul or many tea party activists.

"Most of the tea party activists are really unhappy about this," said Oregon delegate Russ Walker. "We feel it's kind of cast a black cloud over the success we had with the platform."

The tea party can be traced to 2008, when voters started protesting spending.

The group broke through nationally in 2009 as opponents to federal legislation expanding access to health care, and then fueled a wave that carried Republicans to enough victories in the House for control.

Now, many tea partiers say the GOP owes them.

"It would have been nice to see some more of a tea-party presence," said Nevada Republican James Smack. "We can't win without them."

Park Chan-wook Sees Brigands

Having won acclaim for the likes of Oldboy, Sympathy For Mr Vengeance and Three? Extremes, director Park Chan-wook has recently made his English-language debut with Stoker. With work on the vampire tale complete, he has selected The Brigands Of Rattleborge as a possible follow-up.

S. Craig Zahler?s Western hit the Black List way back in 2006, and had been cooling its heels in development thanks to its violent content. Park?s interest means it?ll likely start moving forward again.

The plot follows a group of bandits who use a torrential thunderstorm as the ideal cover to rob a small town. What they don?t count on is a local doctor deciding to take revenge on the thieves. Nobody's plan goes smoothly and much blood is spilt. Frankly, it sounds exactly like the sort of material Park could really pull off, though according to Variety it?ll need to lock in a big name or two to get it made and sold.

Stoker, which features Nicole Kidman, Mia Wasikowska, Dermot Mulroney and Matthew Goode, is set to arrive on March 1 next year.

Lytham Hall included on Heritage Open Days list for Lancashire ...

The Victorian dining chairs and antique bookcases of Lancashire?s historic buildings will be under the spotlight from 6th to 9th September, when the doors are flung open for this year?s Heritage Open Days event.

Organised by English Heritage, the four day event allows people free access to properties they would normally be unable to see, or else have to pay an entrance fee for. In Lancashire, this means the antique dining tables of Gawthorpe Hall, the Georgian silverware of the Lancashire Regiment, the exquisite interiors of Croston Old School and other such attractions will all be on view to the non-paying public.

Naturally, this being Lancashire, many of the events will have an industrial slant, with venues like Bancroft Mill and the Old Mount Engine House puffing away in the otherwise peaceful Ribble Valley. antique desk fans, however, will head for cultural hotspots like Lytham Hall, an 18th Century stately home on the site of a 12th Century manor house and priory. Built for the flamboyant Clifton family, the estate originally covered 16,000 acres and was totally self-supporting ? it even had its own brewery and race-course. Normally, tours of the house ? which is currently undergoing restoration ? are by prior arrangement only, but from 6th to 9th September the family portraits, Georgian silverware and antique marquetry furniture of this Lancashire family will be available to all.

Lytham Hall ceased being a private dwelling in the 1960s, yet still retains the feel of a family home, and its Victorian dining chairs and antique dining tables are used by Lancashire wedding parties on a regular basis. An antique dealer in an area like Preston will have similar fine furniture for sale.

So it has been a while since I've blogged on the Scheffilm website. ?Apologies for this as we have been busy preparing to move interstate and a few projects are taking number one?priority at the moment.

New Bluetooth standards good news for fitness devices

An update to the Bluetooth standard is making it easier for devices like Nike+ and the Fitbit, which need to transmit information like steps taken and pedal cadence, ?but had to brew their own solutions for doing so. Now they can use Bluetooth, just like earpieces and other accessories,?to do so.

These popular fitness accessories have been working with juryrigged solutions over the last couple years, which isn't a problem for a big company like Nike to tackle, but a startup on a shoestring budget doesn't have the cash or time to build their own wireless protocol.

The "Bluetooth Smart"?update will allow this type of simple, real-time data flow to happen over a device's?ubiquitous wireless interface that has, in the past, been used almost entirely for audio. It should make it easier for new devices to work with smartphones to do interesting things like combine a map of your bike ride?with your pace.

There aren't any Bluetooth Smart devices yet, but expect them to show up soon, perhaps at the upcoming CES consumer electronics show in January. More information on the new interface can be found in the Bluetooth Special Interest Group's press release.

India's Gandhi slams opposition MPs over deadlock

Sonia Gandhi, the president of India's ruling Congress party, on Tuesday upped the stakes in a six-day parliamentary stand-off that has paralysed government plans to revive the slowing economy.

Gandhi, the grand matriarch of Indian politics, attacked the main opposition Bharatiya Janata Party (BJP) for holding noisy protests every day in parliament that have forced business to be abandoned.

The BJP is demanding that Prime Minister Manmohan Singh resign over allegations that his government missed out on billions of dollars of revenue by giving away coal rights.

"We are ready to face the challenges before us," Gandhi, wife of former prime minister Rajiv Gandhi, told a Congress party meeting. "Let us stand up and fight, fight with a sense of purpose and fight aggressively."

She said the BJP's protests in both the upper and lower chamber were a form of "blackmail" and accused the opposition of having "scant respect" for democracy.

On Monday, Premier Singh, 79, was howled down by opposition MPs as he tried to defend himself over a report by the official auditor that said mining rights were handed out in a process that "lacked transparency and objectivity".

Since he was re-elected in 2009, Singh has struggled with falling economic growth, damaging corruption scandals and accusations of policy drift after U-turns on key reform plans.

"Our focus... must be to revive and sustain the momentum of high economic growth," Gandhi told her party.

"Dialogue, debate and discussion is the only way forward... but the opposition has to come forth and show a sense of responsibility."

India recorded near double-digit expansion over much of the last decade but the economy grew by just 5.3 percent in the January-March quarter, a rate that threatens to stall the country's transformation since the early 1990s.

The BJP on Tuesday dismissed Gandhi's criticism and again forced parliament to be adjourned for the day as its members stood up, waving papers and shouting for Singh's resignation.

"We admit that parliament is a place for debate, (but) it is also a place for accountability," BJP spokesman Ravi Shankar Prasad said.

The BJP has regularly disrupted parliament since the last election, with protests over the government's alleged mis-selling of mobile phone licences wiping out the entire 2010 winter session.

?We humans are very sensitive to others? mistakes,? Masaki Isoda of the Okinawa Institute of Science and Technology in Japan told Science News. And the same is true for other animals, his new data show.

Isoda?s team has discovered that in monkeys, a small part of the animal?s brain is activated when a companion monkey makes an error. The finding appeared August 5 in a scientific journal.

TAMPA, Fla. (AP) ? On the eve of their national party conventions, Barack Obama and Mitt Romney are locked in a close race to amass the requisite 270 Electoral College votes for victory. And the contest is exactly where it was at the start of the long, volatile summer: focused on seven states that are up for grabs.

Neither candidate has a significant advantage in Colorado, Florida, Iowa, Ohio, Nevada, New Hampshire and Virginia, which offer a combined 85 electoral votes, according to an Associated Press analysis of public and private polls, spending on television advertising and numerous interviews with Republican and Democratic strategists in battleground states.

The analysis, which also took into account the strength of a candidate's on-the-ground organization and travel schedules, found that if the election were held today, Obama would have 19 states and the District of Columbia, offering 247 votes, solidly in his column or leaning his way, while Republican Romney would have 24 states with 206 votes.

Obama won all seven of the too-close-to-call states in 2008, and they are where the race will primarily be contested in the homestretch to the Nov. 6 election.

Romney also is considering trying to expand the battleground map in the coming weeks, watching for an opportunity to compete more aggressively in Wisconsin, and possibly Michigan and Pennsylvania. GOP-leaning groups have been running ads in those states to lay the groundwork for Romney.

Ten weeks before Election Day, the AP analysis isn't meant to be predictive but rather is intended to provide a snapshot of a race that's been stubbornly close all year.

Among the unknowns that could shake up the electoral landscape before November: the latest unemployment figures that come out early next month, an unexpected foreign policy crisis in Syria or Iran and the outcome of the candidates' October debates.

Both sides are working to persuade the 23 percent of registered voters who said in an Associated Press-GfK poll that they are either undecided about the presidential race or iffy in their support for a candidate.

Too woo them, the campaigns and political parties, along with allied groups with access to unlimited financial contributions, have already spent an astounding $540 million on television advertising, according to ad spending reports provided to the AP. And there's more to come.

Over the past three months, the campaign took a sharply negative turn, at times becoming nasty and personal.

Obama sought to define Romney early as a ruthless corporate raider for his time at the head of a private equity firm in Boston, and as an out-of-touch rich man keeping secrets about his wealth. Romney, in turn, worked to cast Obama as a failed president on a host of fronts, primarily the economy.

Both candidates have hit road bumps: Obama saw the unemployment rate rise to 8.3 percent and gave Republicans an opening to argue that he was unfriendly to small business. Romney had a widely panned foreign trip and made a series of potentially problematic comments, most recently joking about the debunked conspiracy theory regarding Obama's citizenship.

The national party conventions, starting with Republicans here on Monday and ending with Democrats the following week in Charlotte, N.C., will set the parameters of the fall campaign, and could provide each side with at least a temporary surge of support in national, if not battleground state, polls.

While Obama has a clear advantage given his incumbency, Romney does have a path to victory ? though it's a steep climb.

He must win most of the seven most competitive states ? Colorado, Florida, Iowa, Ohio, Nevada, New Hampshire and Virginia ? in order to reach the magic number. For instance, he can lose Ohio's 18 electoral votes and still become president if he wins the other six and hangs onto those already in his grasp. It's difficult to see a scenario where Romney wins without a victory in Florida, which offers 29 electoral votes.

Neither side expects a dramatically different playing field this fall.

"You know the states that are in play," said Obama's campaign manager Jim Messina. "I don't think there's going to be a surprise."

Romney's political director Rich Beeson makes the same point: "I don't think you're going to see the map go crazy."

Still, once their conventions are over, both campaigns will commission polls in the hardest-fought states to determine whether to shift their strategies. The candidates and their allied outside groups will pull money and manpower from states that are moving out of reach while relocating it to others they may now think they have a shot at winning.

"We're in a holding pattern," said Charlie Black, a veteran Republican campaign strategist and informal adviser to Romney.

Perhaps not for long.

With a huge cash advantage, Romney is considering trying to put more states in play ? and creating more state-by-state paths to reach 270. He's closely watching to see whether it's worth it to compete aggressively in Wisconsin, now that native son Rep. Paul Ryan is on the ticket.

The Republican National Committee and GOP allies have been advertising in the state in hopes of making it competitive; at least one poll shows they've had some success and the race appears close. Obama, who has a formidable campaign on the ground that includes the state's active labor and minority blocs, hasn't advertised there but might be forced to do so.

Romney also is eyeing a deeper investment in Michigan, where he campaigned Friday, and Pennsylvania, where Ryan was last week. Obama carried both states in 2008, but the GOP sees promise in the economically struggling northern industrial states, especially among working-class, white voters.

The Republican may have the money to expand the map.

August financial reports show that Romney's overall fundraising apparatus ? his campaign, the RNC and a separate joint-fundraising committee ? had roughly $177 million in the bank at the end of July. The reports are the most recent public data.

And to a greater degree than Obama, Romney also has amassed an untapped stockpile of general election money that he plans to use this fall. He can begin spending it immediately upon accepting the nomination for president at the convention's close Thursday night.

Obama and his comparable committees, in turn, had only about $127 million on hand, according to the most recent report.

He also must wait until he accepts his party's nomination on Sept. 6, the close of the Democratic convention, to start spending his general election money.

Unlike Romney, Obama isn't focused on expanding the map in earnest.

He's mostly looking to hang onto as many of the states he won four years ago, with Ohio being of particular focus. In recent months, Obama's standing there has strengthened, the unemployment rate has dropped and last week General Motors announced a $200 million expansion of a northeast Ohio plant to continue building the Chevrolet Cruze there.

Beyond playing defense, Obama's team is watching to see whether the political terrain becomes more favorable to him in Missouri in the aftermath of controversial abortion and rape comments by Rep. Todd Akin, a GOP Senate candidate.

The backlash has been fierce, and polls show Democratic Sen. Claire McCaskill ? arguably the most vulnerable Senate Democrat ? having gained ground. Obama competed for Missouri four years ago but lost and hasn't run ads there this year. Romney has had a comfortable advantage there.

If they have the money to do it, both sides will engage in head-faking: spending money in places simply to force the other side to defend their home turf. For example, if Romney goes after Pennsylvania, which has voted Democratic in all recent presidential elections, Obama would likely have to spend money to defend it, limiting the amount of cash he'll have available to spend in more competitive states, like Florida or Virginia.

Smiling Norway mass killer Breivik jailed for 21 years

An Oslo court on Friday found Anders Behring Breivik guilty of "acts of terror" and sentenced him to 21 years in prison for his killing spree last year that left 77 people dead.

The five judges unanimously found Breivik sane, a verdict in line with what the far-right extremist himself wanted, bringing to an end a spectacular trial for the attacks that traumatised normally tranquil Norway and shocked the world.

Breivik killed eight people in an Oslo blast and took 69 more lives, mostly teenagers', in a shooting frenzy at an island summer camp on July 22, 2011.

"The ruling is unanimous," presiding judge Wenche Elizabeth Arntzen told the court.

"He is sentenced to prison for 21 years, with a minimum of 10 years," she added. Under Norwegian law the sentence could be extended.

Breivik, wearing a dark suit with a white shirt and a grey tie, smiled as the verdict was read out in court.

Survivors of the Utoeya island massacre took to Twitter immediately to comment on the sentencing, with Emma Martinovic tweeting: "YEEEEEEESSSSSSSS!!!"

And Viljar Hansse, who took a bullet to the head in the massacre, tweeted: "Finished. Period."

Breivik has previously said he would not appeal a prison sentence, as he wanted to be found sane so his Islamophobic ideology would not be considered the rantings of a lunatic.

Norway's penal code does not have the death penalty or life in prison, and the maximum prison term for Breivik's charges is 21 years. However, inmates who after that are still considered a threat to society can be held indefinitely.

The 33-year-old loner, who made a right-wing salute in court after his handcuffs were taken off, had confessed to the attacks, seeing himself as a Nordic warrior against Europe's "Muslim invasion" and against all those who promote multiculturalism.

The main question the court had to determine was whether he was sane and could be held responsible for his actions.

Ironically, the prison sentence is what Breivik, most of the families of the victims had wanted, and the general public in Norway wanted.

But Prosecutor Svein Holden had called for him to be sentenced to closed psychiatric care, arguing that "it would be worse to sentence someone who is psychotic to prison than to send someone who is not psychotic to psychiatric care."

Breivik, who laid out his hateful world view in a rambling 1,500-page online manifesto, was diagnosed with paranoid schizophrenia and declared criminally insane after his bloody rampage on July 22, 2011.

However, a public outcry led to a second assessment which found him legally sane -- a view shared by most Norwegians in polls, and by Breivik himself who has said he would accept prison but appeal against closed psychiatric treatment.

Prosecutor Svein Holden, who wants Breivik to be found criminally insane, has said that "it would be worse to sentence someone who is psychotic to prison than to send someone who is not psychotic to psychiatric care."

Breivik hopes to speak again on the final day of his trial, at which Norway has gone to great lengths to stress its free and fair judicial process.

In previous testimony during the 10-week trial that ran until June, Breivik laid out in chilling detail what motivated him to meticulously plan for years and then execute Norway's worst massacre since World War II.

Breivik has called himself a "foot soldier" for the 'Knights Templar', allegedly a clandestine ultra-right group named after an order of Christian Crusaders of the Middle Ages. Police doubt the group's existence.

He has also railed against "cultural Marxists" whose support for immigration he blames for the emergence of a "Eurabia", the reason why he targeted the centre-left government and a summer youth camp run by the Labour Party.

The court heard how Breivik spent years planning the bloodbath, using a farm as cover for purchasing the chemical fertilizer he used for the almost one-tonne bomb he set off in a rented van outside Oslo's main government building.

In his years of seclusion, Breivik said he practised meditation, worked out and used steroids to steel his mind and body, while playing video shooting- and role-playing games for relaxation.

He joined a pistol club and obtained a hunting licence to get the 9mm Glock handgun and Ruger semi-automatic rifle which he used to mow down terrified young people, the youngest just 14, trapped on the tiny lake island of Utoeya.

Dressed in a police uniform, he methodically shot dead 67 people, many at point blank range, and two more died as they fell to their deaths or drowned while trying to escape the more than hour-long shooting spree.

During his trial Breivik showed little emotion and no remorse and once described his mass slaughter as "cruel but necessary" to protect Norway from multiculturalism. At one stage he told the court: "I would do it again."

Researchers at Moffitt Cancer Center and colleagues have identified PHF20, a novel transcriptional factor, and clarified its role in maintaining the stability and transcription of p53, a gene that allows for both normal cell growth and tumor suppression. PHF20, the researchers found, plays a previously unknown and unique role in regulating p53.

When p53 is activated, it can mend DNA damage and eliminate cancer cells by binding to DNA. How p53 maintains its basal level and becomes activated remain elusive, but identifying transcription factor PHF20 and understanding its interaction with p53 and its induction of p53 protein stability and transcription has provided a clue.

Results of their research appeared in a recent issue of Nature Structural & Molecular Biology and also in The Journal of Biological Chemistry.

"When a cell undergoes alterations that predispose it to become cancerous, p53 is activated to either mend the DNA damage or eliminate the affected cells, thereby preventing the development of tumors," said Jin Q. Cheng, Ph.D., M.D., a senior member of the Molecular Oncology Department and Molecular Oncology and Drug Discovery Program at Moffitt. "A number of mechanisms normally keep a regulatory strong check on p53 and allow for rapid activation. Still much is unknown about the mechanism of p53 regulation."

After identifying PHF20 as a novel transcriptional factor, the researchers set out in subsequent studies to probe the function of human PHF20 and its effect on p53. They found that PHF20 not only transcriptionally induces p53 but also directly interacts with and stabilizes p53. Akt negatively regulates these processes by interaction and phosphorylation of PHF20.

To determine whether the absence of PHF20 might regulate stress-induced p53 expression, the researchers "knocked down" PHF20. In doing so, they demonstrated that in the absence of PHF20, p53 was reduced. These findings established the role of PHF20 as a key regulator of p53 and additional link between Akt and p53.

According to Cheng, the identification of PHF20 as a regulator of p53 is significant because PHF20 "participates in simultaneous multiple interactions with other proteins and DNA" and serves to stabilize and induce p53.

"Regulation of p53 is critical to allow both normal cell growth and tumor suppression," explained Cheng. "However, further investigation is required to understand PHF20 tumor suppressor function and its possible involvement in human malignancy."

We're all too familiar with JavaScript tests like Sunspider -- we use them all the time to gauge browser speeds on phones and tablets. Most of these have little direct correlation to the sites we're visiting on our devices, however, and seldom acknowledge that we're testing with anything but a desktop. Google wants to drag the experience into this decade with its new Octane benchmark. The collection of tests uses real-life examples of JavaScript code -- who knew web-based Game Boy emulators were so popular? -- to generate results for a simple, more-is-better scoring system. Its interface also scales dynamically and should fit just about any screen size. While we can't guarantee that Octane will find a place within our own battery of tests, both the benchmark itself and the source code are available for anyone to investigate.
